To encounter the boss, you must complete a lengthy Levaine’s side quest chain in the Carcadia hub, containing a total of five quests, including Enter the Electi I, Enter the Electi II, the Council Divided, a Traitor Within, and the Mole’s Gambit. Afterward, you can encounter the boss and defeat him to collect the Rainbow Vomit from his drop.
Since the legendary shotgun does not appear on the very first try, you need to farm the boss using Moxxi’s Big Encore machine.
