The chest is located at the heart of the fort, but getting to it won’t be a simple stroll. You’ll need to either sneak past the guards or take them down, depending on your preferred gameplay.
Once you’ve entered the center of the fort, Naoe’s kunai will prove useful in that it lets you quietly and quickly kill enemies from afar. Be sure to watch for a Samurai Daisho in the area. This enemy is a big presence, making a head-on attack a risky endeavor, so be stealthy. Alternatively, you can just use Yasuke and go in and fight everyone you see.
